The basic principle of HIC is that proteins have hydrophobic areas that bind to the beads in HIC columns containing high salt concentrated binding buffers. Essentially HIC purifies proteins based on their hydrophobicity. The salt in the binding buffer solution in the HIC column reduces solvation of the GFP which is what causes the hydrophobic regions to stick to the beads in the column. When the salt concentration is decreased with the elution buffer the proteins no longer stick as strongly to the beads and drip through to be isolated. 3. In preparing cells for chromatography it is important to add lysozyme because it digests bacterial cell wall which allows the GFP in the cells to be released into the solution so that it can then be isolated in the HIC process. The freezing step that follows is also important because it causes the bacteria to explode and rupture completely so it just further ensures that the GFP is released. 4. Tube 1 was clear because the binding buffer with a high salt concentration mixed with supernatant caused the hydrophobic areas in the proteins to stick to the beads so that the only thing that ran through the HIC column was the hydrophilic proteins. The second tube was clear as well because it was another step to help wash the extra hydrophilic proteins through with the addition of the wash buffer to the column. The last tube was the only tube that glowed because elution buffer was added to the column which caused the salt concentration to decrease and the hydrophobic proteins stuck to the beads in the column less strongly and dripped through. Ask your professors about these. Take a really close look at past work, and at the mark it got: see if you can figure out why – and what you could do to improve your own work. Astronaut Neil Armstrong stepped out of the lunar lander Eagle, descended a ladder, and set foot on the surface of the Moon. Then, he spoke the most famous words of the 20th Century: Its one small step for man, one giant leap for mankind. His action  was the culmination of years of research and development, success and failure, all sustained by both the U.S. and then-Soviet Union in the race to the Moon. After his high school years at Blume High School in Wapakonetica, Armstrong decided to pursue a degree in aeronautical engineering from Purdue University before committing to serving in the Navy.   In 1949, Armstrong was called to Pensacola Naval Air Station before he could complete his degree. There he earned his wings at the age of 20, the youngest pilot in his squadron. He flew 78 combat mission in Korea, earning three medals, including the Korean Service Medal. Armstrong was sent home before the conclusion of the war and finished his bachelors degree in 1955. Testing New Boundaries After college, Armstrong decided to try his hand as a test pilot. He applied to  National Advisory Committee for Aeronautics (NACA) - the agency that preceded NASA - as a test pilot, but was turned down. So, he took a post at Lewis Flight Propulsion Laboratory in Cleveland, Ohio. However, it was less than a year before Armstrong transferred to Edwards Air Force Base (AFB) in California to work at NACAs High Speed Flight Station. During his tenure at Edwards Armstrong conducted test flights of more than 50 types of experimental aircraft, logging 2,450 hours of flight time. Among his accomplishments in these aircraft, Armstrong was able to achieve speeds of Mach 5.74 (4,000 mph or 6,615 km/h) and an altitude of 63,198 meters (207,500 feet), but in the X-15 aircraft. Armstrong had a technical efficiency in his flying that was the envy of most of his colleagues. However, he was criticized by some of the non-engineering pilots, including Chuck Yeager and Pete Knight, who observed that his technique was too mechanical. They argued that flying was, at least in part, feel, that it was something that didnt come naturally to the engineers. This sometimes got them into trouble. Neil Armstrong was a test pilot before coming to NASA. This shows him at the Dryden Research center in 1960 after he became a NASA research test pilot. He flew missions in the first X-15 rocket plane. NASA   While Armstrong was a comparatively successful test pilot, he was involved in several aerial incidents that didnt work out so well. One of the most famous occurred when he was sent in an F-104 to investigate Delamar Lake as a potential emergency landing site. After an unsuccessful landing damaged the radio and hydraulic system, Armstrong headed toward Nellis Air Force Base. When he tried to land, the tail hook of the plane lowered due to the damaged hydraulic system and caught the arresting wire on the airfield. The plane slid out of control down the runway, dragging the anchor chain along with it. The problems didnt end there. Pilot Milt Thompson was dispatched in an F-104B to retrieve Armstrong. However, Milt had never flown that aircraft and ended up blowing one of the tires during a hard landing. The runway was then closed for the second time that day to clear the landing path of debris. A third aircraft was sent to Nellis, piloted by Bill Dana. But Bill almost landed his T-33 Shooting Star long, prompting Nellis to send the pilots back to Edwards using ground transportation. Crossing Into Space In 1957, Armstrong was selected for the Man In Space Soonest (MISS) program. Then in September 1963, he was selected as the first American civilian to fly in space.   Three years later, Armstrong was the command pilot for the Gemini 8 mission, which launched March 16. Armstrong and his crew performed the first-ever docking with another spacecraft, an unmanned Agena target vehicle. After 6.5 hours in orbit they were able to dock with the craft, but due to complications, they were unable to complete what would have been the third-ever extra-vehicular activity, now referred to as a spacewalk. Armstrong also served as the CAPCOM, who is typically the only person who to communicate directly with the astronauts during missions to space. He did this for the Gemini 11 mission. However, it was not until the Apollo program began that Armstrong again ventured into space. The Apollo Program Armstrong  was  commander of the backup crew of the Apollo 8 mission, though he had been originally scheduled to back-up the Apollo 9 mission. (Had he remained as the  backup commander, he would have been slated to command Apollo 12, not  Apollo 11.) Initially, Buzz Aldrin, the  Lunar Module Pilot, was to be the first to  set foot on the Moon. However, because of the positions of the astronauts in the module, it would require Aldrin to physically crawl over Armstrong to reach the hatch. As such, it was decided that it would be easier for Armstrong to exit the module first upon landing. Apollo 11 touched down on the surface of the Moon on July 20, 1969, at which point Armstrong declared, Houston, Tranquility Base here. The Eagle has landed. Apparently, Armstrong had only seconds of fuel left before the thrusters would cut out. If that had happened, the lander would have plummeted to the surface. That didnt happen, much to everyones relief. Armstrong and Aldrin exchanged congratulations before quickly preparing the lander to launch off the surface in case of an emergency. Humanitys Greatest Achievement On July 20, 1969, Armstrong made his way down the ladder from the Lunar Lander and, upon reaching the bottom declared Im going to step off the LEM now. As his left boot made contact with the surface he then spoke the words that defined a generation, Thats one small step for man, one giant leap for mankind. NASA   About 15 minutes after exiting the module, Aldrin joined him on the surface and they began investigating the lunar surface. They planted the American flag, collected rock samples, took images and video, and transmitted their impressions back to Earth. The final task carried out by Armstrong was to leave behind a package of memorial items in remembrance of deceased Soviet cosmonauts  Yuri Gagarin  and Vladimir Komarov, and  Apollo 1  astronauts Gus Grissom, Ed White and  Roger Chaffee. All told, Armstrong and Aldrin spent 2.5 hours on the lunar surface, paving the way for other Apollo missions. The astronauts then returned to Earth, splashing down in the Pacific Ocean on July 24, 1969. Armstrong was awarded the Presidential Medal of Freedom, the highest honor bestowed upon civilians, as well as a host of other medals from NASA and other countries. The Main Sources Of Carbon Dioxide Emissions Environmental Sciences Essay Since the Industrial Revolution, human activities such as the burning of oil, coal and gas, as well as deforestation have greatly increased CO2 concentrations in the atmosphere. Almost all CO2 emissions (about 96.5%) come from fossil fuels use. The 3 types of fossil fuels that are used the most are coal, natural gas and petroleum. When fossil fuels are combusted, the carbon stored in them is emitted almost entirely as CO2. The three main sectors that use fossil fuels are: à ¢Ã¢â€š ¬Ã‚ ¢ Transportation à ¢Ã¢â€š ¬Ã‚ ¢ Utilities (power, gas, oil etc) à ¢Ã¢â€š ¬Ã‚ ¢ Industrial production Transportation: The first new projections of future aircraft emissions in 10 years predict that carbon dioxide and other gases from air traffic will become a significant source of global warming as they double or triple by 2050. à ¢Ã¢â€š ¬Ã‚ ¢ The most important source of CO2 emissions worldwide is caused by the transportation of goods and people. Whats worse is that 99% of the energy used to transport people and goods all over the world comes from the combustion of fossil fuels. Industrial production: à ¢Ã¢â€š ¬Ã‚ ¢ Manufacturing and industrial processes all combine to produce large amounts of each type of greenhouse gas but specifically large amounts of CO2 because of two reasons. First, many manufacturing facilities directly use fossil fuels to create heat and steam needed at various stages of production. Second, their energy intensive activities use more electricity than any other sector so unless they are using renewable sources the energy that they use is responsible for vast amounts of emissions. à ¢Ã¢â€š ¬Ã‚ ¢ By industrial production we are mainly talking about manufacturing, construction, mining, and agriculture. Manufacturing is the largest of the 4 and can be broken down into 5 main categories: paper, food, petroleum refineries, chemicals, and metal/mineral products. These categories account for the vast majority of the energy use and CO2 emissions by the sector.3, 4 Land-use Change It is estimated that man-made changes in land-use have, until now, produced a cumulative global loss of carbon from the land. Widespread deforestation has been the main source of this loss, estimated to be responsible for nearly 90 percent of losses since the mid-nineteenth century. Losses primarily occur due to the relatively long-term carbon sinks of forests being replaced by agricultural land. The conversion of land from forested to agricultural land can have a wide range of negative effects as far as greenhouse gas emission is concerned. Soil disturbance and increased rates of decomposition in converted soils can both lead to emission of carbon to the atmosphere, with increased soil erosion and leaching of soil nutrients further reducing the potential for the area to act as a sink for carbon. Respiration Respiration, both on land and in the sea, is a key component of the global carbon cycle. On land, an estimated 60 Pg C (60 billion tones) is emitted to the atmosphere each year by autotrophic respiration. In the sea, autotrophic respiration is thought to account for about 58 Pg of the dissolved inorganic carbon in surface waters each year, with the contribution of heterotrophic respiration being 34 Pg C. Effects of increasing temperatures on pollution can be observed which : 1. Higher temperatures due to carbon dioxide increased the chemical rate of ozone production in urban areas 2. Increased water vapor due to carbon dioxide-induced higher temperatures boosted chemical ozone production even more in urban areas. Engineers have designed a simple, sustainable and natural carbon sequestration solution using algae. A team at Ohio University created a photo bioreactor that uses photosynthesis to grow algae, passing carbon dioxide over large membranes, placed vertically to save space. The carbon dioxide produced by the algae is harvested by dissolving into the surrounding water. The algae can be harvested and made into biodiesel fuel and feed for animals. A reactor with 1.25 million square meters of algae screens could be up and running by 2010. America is by far the largest contributor to global warming than any other country releasing a quarter of the worlds carbon dioxide the primary cause of global warming. Bayless, with a team at Ohio University, created a photo bioreactor that uses photosynthesis to grow algae just like a plant would take carbon dioxide up and, through the energy of the sun, convert that into oxygen. That passes the carbon dioxide over these membranes, Ben Stuart, an Ohio University environmental engineer, tells DBIS. These membranes are fabric just like your shirt. Its a woven material, and as the carbon dioxide pass by them, that carbon dioxide dissolves into the water. That carbon dioxide is broken down by the algae. Nitrogen and clean oxygen are released back into the atmosphere. But to capture the CO2 created from a power plant, algae would have to fill a building the size of Wal-Mart. The size of these things would be enormous, about an acre worth of land space. And so the flu gases would run through this huge building and the algae would be growing on the suspended vertical surfaces. Stuart says. But what makes it cost effective? The customer in this case is the auto makers who are intended to buy the newly developed compact disc player to fix them in the automobiles. Bert (1998) explains that the best channel that any manufacturer should prioritize on is one that makes the end product user happiest, so that they are always willing to buy again the same product from the same channel member. Waiting time and delivery time:-this refers to the length of time that the customer waits, for the receipt of goods. Customers always prefer delivery channels that are fast and efficient. Fast delivery of services requires output levels of great services (Kotler, 2000). The CD player manufacturer needs to engage a channel member that provides fast services at convenient time to the customer. Spatial convenience:-This expresses the extent to which the marketing channel simplifies the purchasing of the products by the customers (kotler, 2000). Customers will always prefer to purchase the products which do not involve so many formalities and that are easy to buy (Bert, 1998). The CD manufacturer should involve a channel that meets this requirement. Customers always prefer a marketing channel that offers grater assortment breath because this will most likely meet their exact needs (Kotler, 2000). In regard to this the CD Player manufacturer should consider also incorporating an installation kit alongside this product so that the customer buys the whole package from one place.
